Kisspeptin-10 is a decapeptide derived from the C-terminus of kisspeptin, the product of the KISS1 gene. Research models show it is the most potent known endogenous agonist of KISS1R (GPR54) and acts as the master upstream regulator of hypothalamic GnRH neurons — the gatekeeper of the entire HPG axis. Loss-of-function mutations in KISS1R cause hypogonadotropic hypogonadism in humans, and exogenous kisspeptin triggers robust LH/FSH surges in clinical investigational studies across both sexes.
